Noggin屬于一組可擴(kuò)散蛋白質(zhì),它們與TGF-α家族的配體結(jié)合,并通過(guò)抑制它們進(jìn)入信號(hào)受體來(lái)調(diào)節(jié)其活性。Noggin最初被確定為BMP-4拮抗劑,其作用對(duì)于頭部和其他背部結(jié)構(gòu)的正常形成至關(guān)重要。因此,Noggin已被證明可以調(diào)節(jié)其他BMP的活性,包括BMP-2,-7,-13和-14。小鼠中Noggin的靶向缺失導(dǎo)致產(chǎn)前死亡和隱性表型顯示嚴(yán)重畸形的骨骼系統(tǒng)。相反,在成熟成骨細(xì)胞中過(guò)度表達(dá)Noggin的轉(zhuǎn)基因小鼠顯示出成骨細(xì)胞分化受損,骨形成減少和嚴(yán)重的骨質(zhì)疏松癥。人類爪蟾的氨基酸序列與非洲爪蟾、大鼠和小鼠的氨基酸序列高度同源。
Noggin belongs to a group of diffusible proteins which bind to ligands of the TGF-α family and regulate their activity by inhibiting their access to signaling receptors. Noggin was originally identified as a BMP-4 antagonist whose action is critical for proper formation of the head and other dorsal structures. Consequently, Noggin has been shown to modulate the activities of other BMPs including BMP-2,-7,-13, and -14. Targeted deletion of Noggin in mice results in prenatal death and recessive phenotype displaying a severely malformed skeletal system. Conversely, transgenic mice over-expressing Noggin in mature osteoblasts display impaired osteoblastic differentiation, reduced bone formation, and severe osteoporosis. The amino acid sequence of human noggin is highly homologous to that of Xenopus, rat and mouse.
